In 2001, Steve Wynn shared his ideas for a 45-story resort, initially named Le Rêve. The buildings were built on the site of the former Desert Inn, whose main tower was blown up in October 2001. The remaining structures were demolished in November 2004.

The resort was built by Marnell Corrao Associates for $2.7 billion, making it the largest privately financed construction project in the United States.

Wynn Las Vegas opened in April 2005. The Wynn and Encore hotels are home to WYNN, a quarterly specialty magazine.

The resort's buildings were featured in footage from the movie "2012," and the comedy action film "Paul Blart: Mall Cop 2" was shot entirely in the gambling house. Movie masterpieces filmed on the compound include: "Frank & Lola," "Compression," "Sinatra 100: All-Star Concert at the Grammys," "Garth Brooks' Holiday Edition," and "House."
